A Study on the Utilization of the Enzyme Immunoassay for the Rapid Drug Susceptibility Test of Mycobacterium tuberculosis -Comparison with DNA Hybridization Method-


Abstract
In keeping with the recent development of rapid identification method and effective antituberculous regimens, it is necessary to develop a rapid drug susceptibility testing method for M. tuberculosis. The DNA hybridization assay using rRNA
sequences
unique to M. tuberculosis has been applied to drug susceptibility testing for M. tuberculosis and has been considered to be a sensitive, specific and rapid method. However, it requires special equipment and is expensive. Therefore it is desirable
to
develop a sensitive and specific but simple and cost effective method. In this study, enzyme immunoassay(EIA) has been developed for rapid drug susceptibility testing for M. tuberculosis and was compared with the standard solid media method and
with the
DNA hydridization assay. The results of this assay were found to be comparable to those of both the solid media and the DNA hybridization methods. However the solid media method with Middlebrook 7H10 took 21 to 27 days, and the DNA hybridization
method
took 3 days for the susceptible strains and 7 days for the resistant strains. The EIA method required 7 days for the susceptible strains and 10 days for the resistant strains. In view of the turn around time, cost, ease of performance and
instrumentation, the EIA method is simple and cost effective. It can be adapted as a routine rapid drug susceptibility testing method for M. tuberculosis.
